Deep within a bacterium long studied by scientists, there was a molecule quietly biding its time — neither flashy nor newly engineered, just overlooked. That all changed when researchers from University of Warwick and Monash University uncovered an intermediate compound hidden in the biosynthetic process of a known antibiotic. They found that this intermediate — named pre‑methylenomycin C lactone — is over 100 times more potent against antibiotic-resistant Gram-positive bacteria than its better-known final product.

The bacterium at the heart of the discovery, Streptomyces coelicolor, has been a workhorse in antibiotic science since the 1950s. Rather than mining new species or environments, the researchers decided to revisit the biosynthetic pathways of this familiar microbe — and found that in altering gene components, they unmasked intermediates that no one had tested for antimicrobial activity. “By deleting biosynthetic genes, we discovered two previously unknown biosynthetic intermediates, both of which are much more potent antibiotics than methylenomycin A itself,” said Professor Greg Challis of Warwick.

One of the key findings: pre-methylenomycin C lactone displayed strong antimicrobial activity against such pathogens as Staphylococcus aureus (including MRSA) and Enterococcus faecium (including VRE). Impressively, under experimental conditions where vancomycin – a last-line antibiotic – loses ground, this compound showed no detectable resistance development in Enterococcus.
